Source Antibody: Rabbit polyclonal antibody for both capture and
detection.
Test Format: DAS ELISA
Reactivity: This test system reacted with all WSMV isolates
tested. Reaction of the ELISA is moderately strong. Optical Density
at 405nm is in a range of 0.600 - 1.600 depending on the
virus titer in the samples tested.
Sensitivity: Sensitivity of the ELISA is moderately high.
The virus can be consistently detected in infected plant leaf tissues diluted
at 1:270 - 1:810.
Specificity: There is no obvious cross reaction with healthy
plant tissues. Background is very low on the negative control wells.
Application: The test can be used to detect WSMV in infected
host plants.

Information About the Virus
Name: Wheat Streak Mosaic Virus
Acronym: WSMV
Synonyms: Marmor virgatum, wheat virus 6, wheat virus 7.
Group/Genus: Rymoviruses
Vector: Mites
Transmission: Seed
Main host plants: Wheat, barley, millet, maize, oats, rye.
Viral Particle: RNA virus with filamentous particles, 700 nm
long and 15 nm in diameter.
Virus Infection: Wheat streak mosaic virus (WSMV) infection
causes systemic mosaic symptoms under both warm and cool temperatures.
When wheat is infected in autumn, plants show severe stunting, varying degrees
of necrosis and death of tillers, and reduced seed set and seed weight.
WSMV is transmitted by the wheat-curl mite, which acquires the virus from the
plant only by nymphs, and transmits the virus by both nymphs and adults of the
mite. The virus does not pass from the adult mite to its progeny through
egg. WSMV is transmitted by seed in very low levels.
WSMV is found in Canada, Jordan, Romania, Russia, the USA
and Yugoslavia. WSMV is severe in the south-central part of the United States,
where a host continuum exists for both mite and virus between spring and
autumn-sown crops.
