Test Format:  DAS ELISA                                   

Reactivity:  This test system reacted with all of the isolates of OYDV tested.  Reaction of the ELISA is very strong.  Optical Density at 405nm is in a range of 2.200 - >3.000 depending on the virus titer in the samples tested.

Sensitivity:  Sensitivity of the ELISA is very high.  The virus can be consistently detected in infected plant tissues diluted at 1:2430.

Specificity:  This test shows no cross-reaction with other potyviruses such as DsMV, LMV, LYSV, MDMV, SCMV, PepMoV, PPV, PRSV, PVV, SbMV, SYSV, TEV, TuMV, TVMV, WMV2, and ZYMV. There is no cross reaction found with healthy plant tissues.  Background is low on all of the negative control wells.

Application:  The test can be used to detect OYDV in infected host plants.

Information About the Virus

Name:  Onion Yellow Dwarf Virus
Acronym:  OYDV
Synonyms:  Allium virus 1, Marmor cepae.
Group/Genus:  Potyviruses

Vector:  Aphids, including Green Peach aphid, Myzus persicae, and other aphid species.
Transmission:  Mechanical
Main host plants:  Onion, garlic leek and shallot.

Virus Particle:  Filamentous particles containing RNA genome
Virus Infection:
  Onion Yellow Dwarf Virus (OYDV) infects onion (Allium cepa), shallot (Allium cepa var. ascalonicum), garlic (Allium sativum), and leek (Allium porrum), and causes yield losses in these Allium crops.  Common symptoms of infected onion include leaf striping and plant stunting.  OYDV induces leaf mosaic in garlic and is part of the garlic mosaic virus complex.
