Reactivity:  This test system reacted with all of the isolates or strains of MNSV tested.  Reaction of the ELISA is very strong.  Optical Density at 405nm is in a range of 1.200 - 2.800 depending on the virus titer in the samples tested.

Sensitivity:  Sensitivity of the ELISA is relatively high.  The virus can be consistently detected in infected plant tissues diluted at 1:810.

Specificity:  This ELISA shows no cross-reaction with other carmoviruses, such as carnation mottle virus, hibiscus chlorotic ringspot virus and pelargonium flower break virus. There is no cross reaction found with healthy cucurbit plant tissues.  Background is low on all of the negative control wells.

Application:  The test can be used to detect MNSV in infected leaves and seeds of cucurbits.

Virus Particle:  Isometric particles containing RNA genome, 30 nm in diameter. 
Virus Infection:
  Melon necrotic spot virus (MNSV) occurs in Japan, the Netherlands, the UK, and the USA.  MNSV is transmitted by the chytrid fungus Olpidium radicaleand in soil.  This virus is also transmitted by mechanical sap inoculation.  Naturally, MNSV occurs only in greenhouse melons and cucumbers, and causes severe yield loss.  The symptoms induced by the virus include necrotic spots or large necrotic lesions on the leaves, and necrosis on the stems of greenhouse melons and cucumbers.
