Test Format: DAS ELISA
Reactivity: This test system reacted with all virus isolates
tested. Reaction of the ELISA is moderately strong. Optical Density at 405nm
is in arrange of 1.000 - 1.600 depending
on the virus titer in the samples tested.
Sensitivity: Sensitivity of the ELISA is very high. The virus can be consistently detected in infected plant tissues diluted at 1:2430 - 1:7290.
Specificity: This test shows no cross-reaction with odontoglossum ringspot virus, pepper mild mottle virus and tobacco mosaic virus. A weak cross-reaction (OD 0.200 - 0.400 at 405 nm) was found with ribgrass mosaic virus and tomato mosaic virus. There is no cross reaction with healthy plant cucurbits tissues. Background is very low on all of the negative control wells.
Application: The test can be used to detect CGMMV in infected cucurbit plants.

Viral Particle: Rod-shaped, 300 x 18 nm, .containing RNA genome.
Virus Infection: Cucumber green mottle mosaic virus (CGMMV)
causes leaf mottling, blistering and distortion with stunted growth in cucumber.
Cucumber fruits are usually symptomless. In watermelon, CGMMV induces
slight leaf mottling and dwarfing, but can cause serious internal discoloration
and decomposition of the fruit. The virus is readily transmitted by mechanical
methods such as foliage contact, handling of the plants during cultivation, soil
contamination. It is also transmitted through infected seeds. CGMMV
is found in Europe, Asia and other areas in the world.
