Reactivity:  The test reacted with all ZGMMV isolates tested. Reaction of the ELISA is moderate strong.  Optical Density at 405nm  is in arrange of 0.750 - >2.700 depending on the virus titer in the samples tested.

Sensitivity:  Sensitivity of the ELISA is moderate high.  The virus can be consistently detected in infected plant tissues diluted  at 1:810 - 1:2430.

Specificity:  This test system reacts negatively with tobamoviruses such as CGMMV, KGMMV, ORSV, RMV, SHMV and TMV. This ELISA shows some cross-reactivity with PMMoV and ToMV at low levels (0.300 - 0.400 of O.D.405nm ). The test does not cross-react with other tobamoviruses such as CGMMV, ORSV, RMV and TMV.   There is no cross reaction with healthy plant  tissues such as zucchini or other host plants. Background is low on all of  the negative control wells.

Application:  The test can be used to detect ZGMMV in infected host plants.

Information About the Virus

Name:  Zucchini Green Mottle Mosaic Virus

Vector:  Non-Vector Transmission
Transmission: Virus is transmitted by seeds.
Main natural host plants: Zucchini.

Virus Infection:  Symptoms are expressed in the leaf and infructescence; include stunting, deformation, and reduced vigor; include stunting of all plant parts; include deformation of leaves and fruits. Symptoms in leaves include size reduction and development of patterns or markings. Leaf margin can be chlorotic and changed in color. Leaves with mosaic and mottle have green mosaic and green mottle. Fruit symptoms include malformation, size reduction, and change in texture.
Diagnostically susceptible host species and symptoms: Datura stramonium — necrotic local lesion. Infection is local. Symptoms visible; after 3 days. Local symptoms include necrotic spots. Local lesions found on leaf. Lesions are necrotic. Diagnostically insusceptible host species: Nicotiana tabacum.

Geographical distribution: Geographical distribution of the virus is probably restricted. The virus occurs in South Korea.
