Source Antibody: Rabbit polyclonal antibody for both capture and
detection.
Test Format: DAS ELISA
Reactivity: The test reacted with all VTMoV isolates tested. Reaction of the ELISA is strong. Optical Density at 405nm is in arrange of 0.960 - >3.000 depending on the virus titer in the samples tested.
Sensitivity: Sensitivity of the ELISA is high. The virus can be consistently detected in infected plant tissues diluted at 1:810 - 1:2430.
Specificity: There is no cross reaction with healthy plant tissues such as velvet tobacco and other host plants. Background is low on all of the negative control wells.
Application: The test can be used to detect VTMoV in infected host plants.

Information About the Virus
Name: Velvet Tobacco Mottle Virus
Acronym: VTMoV
Synonyms:
Group/Genus: Sobemovirus
Vector: Transmitted by a vector; an insect; Cyrtopeltis nicotianae:
Miridae. Transmitted in a semi-persistent manner (atypically). Virus does not
multiply in the vector; does not require a helper virus for vector transmission
(Gibb and Randles, 1988).
Transmission: Transmitted by mechanical inoculation; not transmitted by
seed.
Main natural host plants: Nicotiana velutina
Virus Infection: Causes systemic mosaic and leaf malformation. Symptoms
persist.
Diagnostically susceptible host species and symptoms: Nicotiana
clevelandii - chlorotic to necrotic local lesions, then systemic vein
yellowing and mosaic; Nicotiana glutinosa, N. × edwardsonii - mild
systemic mosaic.
