Source Antibody: Rabbit polyclonal antibody for both capture and
detection.
Test Format: DAS ELISA
Reactivity: The test reacted with all SPFMV isolates tested. Reaction of the ELISA is moderate strong. Optical Density at 405nm is in arrange of 0.750 - 2.850 depending on the virus titer in the samples tested.
Sensitivity: Sensitivity of the ELISA is moderate high. The virus can be consistently detected in infected plant tissues diluted at 1:810.
Specificity: There is no cross reaction with healthy plant tissues such as rice and other host plants. Background is relative low on all of the negative control wells.
Application: The test can be used to detect SPFMV in infected host plants.

Information About the Virus
Name: Sweat Potato Feathery Mottle Virus
Acronym: SPFMV
Synonyms: sweet potato chlorotic leaf spot virus, sweet potato internal cork
virus, sweet potato russet crack virus, sweet potato virus A (Hollings et al.,
1976; Sheffield, 1957).
Group/Genus: Potyvirus
Vector: Transmitted by a vector; an insect; Aphis gossypii, A.
craccivora, Lipaphis erysimi, Myzus persicae; Aphididae. Transmitted in a
non-persistent manner.
Transmission: Virus transmitted by mechanical inoculation; transmitted by
grafting; not transmitted by contact between plants; not transmitted by seed;
not transmitted by pollen.
Main host plants: Ipomoea spp.
Virus Infection: Symptoms disappear soon after infection (except for
russet crack strain symptoms which persist on roots). vein clearing, vein
feathering, chlorotic spots, in cv. Jersey also causes root necrosis if infected
by russet crack strain.
Diagnostically susceptible host species and symptoms: Ipomoea incarnata,
I. nil - systemic vein clearing, vein banding, ringspots. I. setosa -
vein clearing followed by remission.
