Test Format: DAS ELISA
Reactivity: The test reacted with all RYMV isolates tested.
Reaction of the ELISA is very strong. Optical Density at 405nm
is in arrange of 1.100 - >3.000 depending
on the virus titer in the samples tested.
Sensitivity: Sensitivity of the ELISA is very high. The virus can be consistently
detected in infected plant tissues diluted at 1:810 - 1:2430.
Specificity: There is no cross
reaction with healthy plant tissues such as rice and other host
plants. Background is
very low on all of the
negative control wells.
Application: The test can be used to detect RYMV in infected
host plants.

Information About the Virus
Name: Rice Yellow Mottle Virus
Acronym: RYMV
Synonyms:
Group/Genus: Sobemoviruses
Vector: Transmitted by a vector; an insect; Sesselia pusilla,
Chaetocnema pulla, Trichispa serica and Dicladispa viridicyanea;
Coleoptera. Transmitted in a semi-persistent manner. Virus lost by the vector
when it moults; does not multiply in the vector; not transmitted congenitally to
the progeny of the vector; does not require a helper virus for vector
transmission.
Transmission: Virus transmitted by mechanical inoculation; not
transmitted by seed.
Main host plants: Oryza sativa, Oryza longistaminata
- chlorosis and mottling.
Virus Infection: Cause symptoms of yellow or orange leaves,
stunting, reduced tillering and sterility, or chlorosis and mottling.
Symptoms persist.
Diagnostically susceptible host species and symptoms: Oryza sativa -
chlorotic dotting of leaf bases, elongating parallel to veins to produce
mottling, stunting; Oryza barthii - yellow spots and vein streaking;
Oryza punctata - systemic chlorotic dots.
Geographical distribution: Spreads in Burkina
Faso, Cote d'Ivoire, Kenya, Liberia, Mali, Niger, Nigeria, Sierra Leone, and
Tanzania (and, possibly, Zanzibar).
