Source Antibody: Rabbit polyclonal antibody for both capture and
detection.
Test Format: DAS ELISA
Reactivity: The test reacted with all BBSV isolates tested. Reaction of the ELISA is moderate strong. Optical Density at 405nm is in arrange of 0.800 - >2.200 depending on the virus titer in the samples tested.
Sensitivity: Sensitivity of the ELISA is moderate high. The virus can be consistently detected in infected plant tissues diluted at 1:810 - 1:2430.
Specificity: There is no cross reaction with healthy plant tissues such as bean, pea, cowpea or other host plants. Background is low on all of the negative control wells.
Application: The test can be used to detect BBSV in infected host plants.

Information About the Virus
Name: Broad Bean Stain Virus
Acronym: BBSV
Synonyms: virus de la mosaique de la fève, MF virus, broad bean F1 virus
(Gibbs et al., 1968), broad bean Evesham stain virus
Group/Genus: Comovirus
Vector: Transmitted by a vector; an insect; Apion vorax, Sitona
spp.; Coleoptera.
Transmission: Virus transmitted by mechanical inoculation;
transmitted by grafting; transmitted by seed (up to 10% in some Vicia faba
cultivars).
Main natural host plants: Vicia faba
Virus Infection: Symptoms include systemic mottling,
necrosis and malformation. Symptoms vary cyclically over a few weeks, or vary
seasonally.
Diagnostically susceptible host species and symptoms: Phaseolus
vulgaris cv. Tendergreen - systemic mottle; Pisum sativum, Vicia faba
- systemic mottle; necrosis.
Geographical distribution: Spreads in the African region and the Eurasian region. Found, but with no evidence of spread, in Australia from imported seed.
